I have a yearling colt who has a homozygeous for tobiano sire and an appie dam.

Could he be homozygeous for tobiano?
 
The colt could only be homozygous for tobiano if the dam also carries the tobiano gene and passed it on to him. Now, she might not show it, but she would have to have it, and it would have to come from one of HER parents. The colt will have one copy of the tobiano gene if his sire was homozygous, so he will be a tobiano pinto, even if he doesn't show it.

If you KNOW the dam does not carry tobiano, then save your money. The foal cannot be homozygous for pinto (tobiano) unless BOTH parents carry tobiano. Is there a particular reason you are wanting to test? I mean does she have small spots and you are wondering if they are coming from the appy or the pinto so you are testing to try and find an answer? If so, even non-homozygous pintos can have small spots (pawprints).
